When selecting a device in Rufus to write the qubes file to my USB, I am encountering the following issue: the only option for a device is multiple partitions and lists the drive letter for both the unpartitioned volume and the partition I created. Wines cannot run on multiple partitions, correct? Does anyone know how I can get around this? Thanks.

Rufus is probably detecting your single device as having two partitions, labeled with different letters. Try doing a flash drive format, and then do a qubes installation on the USB drive according to the instructions below:
